In the Lab for the Understanding of Network Effects (LUNE), we are discovering the engineering principles of how drug effects are propagated through protein interaction networks. We are developing tools to measure the propagation of disease-modifying effects (related to drug efficacy) and the propagation of unintended side-effects (related to drug safety). We aim to understand the relatedness of safety and efficacy network effects to enable rational selection of new drug targets using an understanding of their potential downstream effects.

As a SPARK fellow and Sanofi iDEA Award recipient, I used protein interaction modeling to understand how proteins downstream of drug targets could associate approved drugs to new diseases. Using this approach, we prioritized drug repurposing opportunities, with an emphasis on discovering treatments for patients suffering from psychiatric and neurological diseases. I developed tools for identifying targets upstream of diseases without existing treatments to advance pathway-driven drug target prioritization.

As an inaugural Fellow with the Center for Excellence in Regulatory Science and Innovation (CERSI), I developed and deployed the PathFX algorithm for understanding proteins downstream of drug targets and how those proteins associated drugs to unintended side effects. The algorithm is currently in use at the US Food and Drug Administration and can be accessed at http://pathfxweb.net
