I continued work started during my PhD on the genetic recoding method to dissect the roles of site-specific protein O-GlcNAcylation. I used CRISPR-Cas9 knock-in technology to site-specifically install S-GlcNAc, a stable analogue of O-GlcNAc, allowing to study the function of the latter.

The aim of my main project was to create a  method to achieve genetic encoding of a stable O-GlcNAc analogue. This involved genetic code expansion via enzyme directed evolution, as well as other innovative protein engineering approaches.

The two side projects were:
1) Biochemical characterisation of a newly discovered intellectual disability mutation in the ogt gene
2) Cell biological and biochemical validation of a novel thio-linked O-GlcNAc transferase inhibitor.
